Contrave is an investigative weight loss drug that has not been approved by the FDA. A mixture of two existing medications, Contrave was designed as a treatment for obesity by Orexigen Therapeutics Inc. The combination is made from naltrexone IR and bupropion SR. Naltrexone is better known as Revia or Vivitrol. Bupropion is well known under as Wellbutrin or Zyban. Naltrexone is typically used to treat drug addition, including common street drugs as well as alcohol. Some of the common side effects that have been reported with Naltrexone have been nausea, constipation, loss of appetite, headaches, anxiety, irritability. Bupropion is an antidepressant which is also used to help people stop smoking. Unlike many common anti-depressants, bupropion is not associated with sexual dysfunction or weight gain.[1]

The FDA will not approve Contrave unless the manufacturer can demonstrate that the drug is not a serious risk to cardiovascular health. In November 2011, Orexigen announced that after months of negotiations with the federal agency, the company may be on a path towards approval.[2]

Common Contrave Side Effects

The full extent of Contrave side effects are not yet known, as the drug has not been approved yet. However, the most common side effects demonstrated in clinical trials included nausea, constipation and headache.[1] Studies also showed that Contrave raised pulse rates and blood pressure slightly, a sign that it could increase the risk of heart attacks, strokes or other cardiovascular problems. Although the participants who used Contrave did not have a higher rate of heart attacks and strokes than those getting the placebo, the FDA ultimately determined that additional research was necessary before the drug could be placed on the market. [3]

Contrave Allergic Reactions

Patients who have demonstrated allergies to either naltrexone or bupropion are likely to experience allergic reactions to Contrave. There also may be inactive ingredients that evoke allergic response. Signs of allergy to Contrave and other medications include rash, hives, or blistering on the skin, which may be accompanied by itching. Cough, nasal leakage and fever can also occur. In rare cases, patients may go into anaphylactic shock, which can be deadly. Signs of this are trouble breathing and hives that occur shortly (usually within an hour) of taking the drug.[4]

Contrave Precautions and Warnings

Contrave has not been approved by the FDA. In denying approval to Contrave in 2010, the FDA cited cardiovascular risks such as heart attack and stroke that do not outweigh the potential benefits of this anti-obesity medication. Additionally, the FDA found Contrave only modestly effective in controlling weight gain. If and when Contrave is approved, full precautions and warnings will be made available to consumers in prescription inserts and online.
